摘要: Lung cancer is one of the most common types cancer, accounting for approximately 15% all cases worldwide. Apoptosis dominant defense mechanism against tumor development. The balance between pro- and antiapoptotic members Bcl-2 protein family can determine cellular fate. venom predatory marine snails Conus estimated to have 100-400 toxins called conotoxins. α-conotoxins known consist selective antagonists nicotinic acetylcholine receptors (nAChRs). cells overexpress several subunits nAChRs are considered as an excellent target new anticancer drugs. We compared cytotoxic effect two synthetic peptides derived from Californiconus californicus, Cal14.1a, Cal14.1b, which only differ by amino acid in their sequence, proapoptotic Bax mRNA expression. determined caspase-3 -7 activation demonstrate apoptosis induction. Results showed that Cal14.1a induces a high Bax/Bcl-2 ratio H1299 (lung cells). Although Cal14.1b has on cells, reducing cell viability 30%, it does not increase ratio, could be explained Glu 15th residue, crucial ability induce apoptosis.
