Production and modulation of interleukin 6 synthesis by synoviocytes derived from patients with arthritic disease.

摘要: Interleukin 6 (IL-6) is a potent cytokine, the biological activities of which include stimulation immunoglobulin secretion, T cell activation, induction acute phase response, activation megakaryocytes, and pyrogenicity. These make it plausible contributor to rheumatoid arthritis. The ability synoviocytes synthesise this potential mediator inflammation was tested. Cultures fibroblast-like cells were established from joint tissue patients with arthritis, degenerative disease, or trauma. Supernatants each diagnostic category contained IL-6-like activity as detected in B9 plasmacytoma proliferation assay. IL-1 stimulated arthritis (n = 5) an average 70,000 U/ml IL-6. Western blot analysis confirmed that these supernatants peptides reacted highly specific antibody A cDNA probe for IL-6 hybridised mRNA derived representative disease state. expression increased by culturing presence 10% calf serum, (30 U/ml), insulin (166 ng/ml), basic fibroblast growth factor (16 ng/ml).
