Concordant Analysis of KRAS Status in Primary Colon Carcinoma and Matched Metastasis

摘要: KRAS somatic mutations are the main predictive factor for non response to EGFR-targeted monoclonal antibodies in metastatic colorectal cancer (mCRC) patients. We compared mutational status primary tumour and corresponding metastases (1 4 sites) 38 mCRC was analysed using direct sequencing, SNAPShot multiplex PCR Scorpion Taqman analysis. Results showed 54% of tumours had mutations. A concordance 97% between primaries sites observed. heterogeneity also demonstrated 5% mCRC. One case with three different harboured mutations, only one represented unique metastasis this patient. concluded there a high metastases. More than informative block more sensitive assay may increase accuracy determination.
