Effect of finite doses of propylene glycol on enhancement of in vitro percutaneous permeation of loperamide hydrochloride.

摘要: We hypothesised that the depletion of propylene glycol from topical formulations applied at clinically relevant doses (approximately mg/cm2) would limit its penetration enhancement effect. The in vitro percutaneous permeation a model drug-loperamide hydrochloride-in containing was therefore investigated under finite dose conditions. flux loperamide and across dermatomed human skin measured. first study examined effect loading gel 12% glycol. second content creams 15 40%. Both studies showed correlation between amount dosed on drug had permeated. substantial relatively small loperamide, strongly suggests, time dependent due to surface not itself. As often experiments do match intended clinical dosage-they are usually much greater-this suggests can be overestimated studies.
