VEGFR1 and VEGFR2 involvement in extracellular galectin-1- and galectin-3-induced angiogenesis.

摘要: Aim:Accumulating evidence suggests that extracellular galectin-1 and galectin-3 promote angiogenesis. Increased expression of and/or has been reported to be associated with tumour progression. Thus, it is critical identify their influence on angiogenesis.Methods:We examined the individual combined effects endothelial cell (EC) growth tube formation using two EC lines, EA.hy926 HUVEC. The activation vascular factor receptors (VEGFR1 VEGFR2) was determined by ELISA Western blots. We evaluated VEGFR1 VEGFR2 levels in endosomes proximity ligation assay.Results:We observed different responses exogenous galectins depending line. An enhanced effect when both were added together. Focusing this effect, we together induced phosphorylation VEGFR2, whereas -3 alone only. In same way, addition a blocking antibody completely abolished increase galectins. contrast, only partially inhibited effect. Finally, decrease endocytic pools, significantly pool. These results suggest action an angiogenesis via activation, which could related receptor endocytosis. © 2013 D'Haene et al.
