摘要: Pneumocystis carinii is an extracellular organism which thought to require attachment alveolar epithelial cells for its growth and replication in humans. Fibronectin (Fn) binding P. essential optimal attachment. This study demonstrates that gp120, a 110-120-kD membrane glycoprotein on carinii, mediates of the cultured lung site Fn carinii. A 51Cr-labeled assay was used quantify cell line A549. Addition free purified from whole organisms, caused significant decrease A549 44.2 +/- 5.5% 22.4 4.2% (P less than 0.01). Preincubation organisms with polyclonal antibody gp120 also resulted marked 46.8% 5.2% 21.3 4.8% Furthermore, addition reduction specific 125I-Fn (from 83.3 8.5 ng 47.1 5.9 ng, P Similarly, anti-gp 120 decreased 74.3 8.4 25.5 5.3 0.001). Solubilized separated by gel electrophoresis blotted demonstrated gp120. In addition, anti-beta 1-integrin antiserum reacted Western blot, suggesting structural homology between beta-subunit integrins. Thus, data suggest functions as protein required cells.
