摘要: The gene expression and enzyme kinetics of acyl coenzyme A:cholesterol acyltransferase (ACAT) were investigated in human monocytes, macrophages, foam cells. Northern blot analysis using a 1.65-kb coding region ACAT cDNA as the probe showed that each cell types exhibited four mRNA transcripts. levels 4.2- 3.7-kb transcripts three- sixfold higher, respectively, macrophages than monocytes. These expressed at same high after conversion to In contrast, 6.3- 4.4-kb for relatively constant level all three types. glyceraldehyde phosphate dehydrogenase, control this study, was also increase accompanied by changes kinetic properties enzyme. Specifically, there 14-fold Vmax 71% decrease Km with respect oleoyl A. Although not definitive, concomitant strongly suggest amount protein increases upon monocytes macrophages. data show can be regulated both substrate expression.
