摘要: In contrast to the murine system, long-term hamster bone marrow suspension cultures maintain proliferation of both pluripotent and committed stem cells in absence an adherent layer without addition exogenous factors, such as hydrocortisone. Addition pokeweed-mitogen-stimulated spleen conditioned medium (SCM) these produces increase number mixed colonies assayed soft-agar, These colonies, which contained four cell lineages--granulocytic, erythroid, megakaryocytic, macrophage--could be generated from grown for over 6 mo. SCM also induces initial rapid expansion myeloid compartment, this results 70% being terminally differentiated granulocytes. contrast, containing hematopoietic produced no change culture. This system allows vitro study process differentiation provides a means examine relationship supernatant populations.
