Maintenance of infectivity of Trypanosoma congolense in vitro with explants of infected skin at 37 °C

摘要: Abstract Glossina morsitans infected with two stocks of Trypanosoma congolense were fed on rabbits and calves to produce local skin reactions containing trypanosomes. Areas removed from the animals used prepare dermal explant cultures in Eagle's MEM RPMI 1640 culture medium, supplemented foetal bovine serum penicillin streptomycin. Cultures incubated at 37 °C media changed 24 48 hour intervals maintain pH 7·0 7·2. There was evidence trypanosome multiplication set up both media; one stock maintained equally well 1640, but other survived better MEM. Explant prepared calf tissues generally yielded more trypanosomes hours than those rabbit tissues. The numbers parasites present near explants for 14 15 days before a decline parasite concentration occurred. organisms retained typical blood stream trypomastigote morphology infective mice periods 21 days. growing primary could not be subpassaged alone or monolayers fibroblast-like cells bovine, murine buffalo origin. Attempts establish by placing directly similar also unsuccessful.
