摘要: DNA ligase activity is strongly accelerated in the presence of high concentrations non-specific polymers, with an accompanying change product distribution characteristics. The rate blunt-end ligation substrate by T4 particularly affected, a shift from closed circular species to linear oligomers. method provides way increase enzymatic and size products. It may be useful when preparing large amounts polymers oligomers or ligating deoxyribooligomers so low concentration that reduced yield would otherwise result.
