摘要: We have tested different cell types as sources for nucleus donors to determine differences in cloning efficiency. When donor nuclei were isolated from cumulus cells and injected into recipient oocytes adult hybrid mice (B6D2F1 B6C3F1), the success rate of was 1.5-1.9%. inbred (C57BL/6, C3H/He, DBA/2, 129/SvJ, 129/SvEvTac), reconstructed did not develop full term or resulted a very low (0-0.3%) with exception 129 strains which yielded 0.7-1.4% live young. fetal (13.5-15.5 dpc), ovarian, testicular used donors, 2.2 1.0% developed offspring, respectively. various somatic (fibroblasts, thymocytes, spleen cells, macrophages) used, receiving thymocyte never beyond implantation, whereas those other did. These results indicate that are necessarily inferior younger (fetal ES cells) context mouse cloning. Although believed less genetic damage than using any low. This may largely be due technical problems and/or genomic reprogramming by rather accumulation mutational cells.
