摘要: Purpose: The current therapeutic approach is not so effective in breast cancer patients. Alternative treatment protocols aimed at different targets need to be explored. We recently reported a novel phosphatidylethanolamine-binding protein, human protein 4 (hPEBP4), as an antiapoptotic molecule. finding led us explore promising for therapy via silencing the expression of hPEBP4. Experimental Design: hPEBP4 clinical specimens was examined by Tissue Microarrays. RNA interference used silence MCF-7 carcinoma cells and effects on cell proliferation, cycle progression, apoptosis, well underlying mechanisms, were investigated. Results: found expressed up 50% cancers but only ser15 , p21 waf/cip Bax, down-regulation Bcl-2 Bcl-xL, which shown depend extracellular signal-regulated kinase 1/2 c-jun NH 2 -terminal activation silencing. Moreover, increased proportion G 0 -G 1 phase observed hPEBP4-silenced TNF-α cyclin A E down-regulated more significantly. Conclusions: effect preferential pattern tissues make new target therapy. Silencing may carcinoma.
