Subventricular Zone-Derived Neuroblasts Migrate and Differentiate into Mature Neurons in the Post-Stroke Adult Striatum

摘要: Recent studies have revealed that the adult mammalian brain has capacity to regenerate some neurons after various insults. However, precise mechanism of insult-induced neurogenesis not been demonstrated. In normal brain, GFAP-expressing cells in subventricular zone (SVZ) lateral ventricles include a neurogenic cell population gives rise olfactory bulb only. Herein, we report evidence that, stroke, these are capable producing new outside bulbs. SVZ labeled by cell-type-specific viral infection method were found generate neuroblasts migrated toward injured striatum middle cerebral artery occlusion. These formed elongated chain-like aggregates similar those SVZ, and chains observed be closely associated with thin astrocytic processes blood vessels. Finally, long-term tracing green fluorescent-labeled Cre-loxP system SVZ-derived differentiated into mature striatum, which they expressed neuronal-specific nuclear protein synapses neighboring striatal cells. results highlight role neuronal regeneration stroke its potential as an important therapeutic target for neurological disorders.
