One-step isothermal RNA detection with LNA-modified MNAzymes chaperoned by cationic copolymer.

摘要: RNA detection permits early diagnosis of several infectious diseases and cancers, which prevent propagation improve treatment efficacy. However, standard technique for such as reverse transcription-quantitative polymerase chain reaction has complicated procedure requires well-trained personnel specialized lab equipment. These shortcomings limit the application point-of-care analysis is critical rapid effective disease management. The multicomponent nucleic acid enzymes (MNAzymes) are one promising biosensors simple, isothermal enzyme-free detection. Herein, we demonstrate simple yet strategies that significantly enhance analytical performance MNAzymes. addition cationic copolymer structural modification MNAzyme enhanced selectivity activity MNAzymes by 250 fold 2,700 fold, respectively. highly simplified system achieved a 73 fM target concentration without additional amplification. robustness in presence non-target was also improved. Our finding opens up route toward development an alternative rapid, sensitive, isothermal, protein-free diagnostic tool, expected to be great clinical significance.
