Role of endocrine, autocrine, and paracrine interactions in the development of mammary hyperplasia in Wnt-1 transgenic mice

摘要: The Wnt-1 proto-oncogene is transcriptionally activated by mouse mammary tumor virus in virus-induced tumors. Previous studies using transgenic mice showed that expression gland causes alveolar hyperplasias which resemble glands of pregnant mice. To understand the role mammogenic hormones genesis these hyperplasias, we examined development before puberty young and effects ovariectomy adrenalectomy on growth morphology hyperplasia. Mammary females hyperplastic as early 1 week after birth. normal structure uterus adult virgin indicated circulating levels ovarian were not elevated. Ovariectomy had no obvious effect hyperplasias. assess possible paracrine stimulation epithelial cells (MEC) stromal cells, transplanted MEC from BALB/c into gland-free fat pads found maintained their ductal without development. Further, did detect mRNA When nude allowed to grow towards existing MEC, remained normal. We concluded hyperplasia solely dependent MEC. speculate may be a factor for only acts locally produce it.
