Stage-specific methylome screen identifies that NEFL is downregulated by promoter hypermethylation in breast cancer

摘要: Aberrant hypermethylation of promoter regions in specific genes is a key event the formation and progression breast cancers, an increasing number marker have been identified. However, few which show methylation change accordance with cancer To identify consistently undergo alterations as tumor develops from benign to malignant form, genome-wide databases cell lines stage I IV were analyzed. Heatmap cluster analysis revealed that changes showed good progression. Seven out 14,495 found be increased alongside level through normal line lines. NEFL, one silico hypermethylated cancer, lower expression MDA-MB-231, well tissues (methylation, p<0.05; expression, p<0.01). The was restored by inducing demethylation MDA-MB-231 cells. Our findings may lend credence possibility using stage-specific patterns biomarkers for estimating prognosis assessing treatment options cancer.
