Phasic effects of glucose, phospholipase A2, and lysophospholipids on insulin secretion.

摘要: Arachidonic acid and lysophospholipids generated by glucose stimulation of phospholipase A2 may be related to the biphasic pattern insulin secretion. Therefore, we examined effects glucose, exogenous A2, lysophospholipids, pharmacological agents which perturb reesterification or oxygenation arachidonic in superfused monolayer cultures neonatal rat islet cells. Nordihydroguaiaretic (20 μm), an inhibitor lipoxygenase, significantly decreased phasic glucose-stimulated secretion, especially during phase 1, suggesting that stimulatory lipoxygenase metabolites contribute stimulation. Treatment with (10 mU/ml) melittin (1.0 2.0 μg/ml) generate de novo caused a monophasic release insulin, followed gradual decline secretion despite continued presence these agonists.
