摘要: PURPOSE. To evaluate the effect of topically administrated latanoprost on optic nerve head (ONH) circulation in Dutch rabbits, cynomolgus monkeys, and normal humans. METHODS. The ONH tissue blood velocity (NB ) was determined using laser speckle method. Latanoprost (0.005%, 30 μl) instilled into one eye, vehicle other eye as a control. In NB measured for 90 minutes after single instillation before 7-day once-daily regimen. 1, 4, 7 days intravenous indomethacin latanoprost-induced change also studied rabbits monkeys. humans, time-course changes were 4.5 hours Intraocular pressure (IOP) systemic parameters simultaneously each experiment. All measurements performed by investigators masked to experimental condition. RESULTS. significantly increased 10% 19% treated eyes (P = 0.035) or regimen 4-day 0.013) whereas there no significant vehicle-treated any experiments > 0.5). Pretreatment with (5 mg/kg) abolished increase but not IOP reduction latanoprost-treated remained unchanged both 0.4), it decreased only latano-prost-treated monkeys < 0.05) humans 0.05). CONCLUSIONS. Topical This independent IOP-reducing probably associated local penetration drug production endogenous prostaglandins.
