摘要: Biomarkers of cancer can indicate the presence disease and serve as therapeutic targets. Our goal is to develop an optical imaging approach using molecularly targeted contrast agents assess several centimeters mucosal surface for mapping expression multiple biomarkers simultaneously with high spatial resolution. The ability image biomarker level heterogeneity in vivo would be extremely useful clinical research, patient selection personalized medicine, monitoring therapy. In this proof-of-concept ex study, we examined correlation neoplasia two clinically relevant biomarkers: epidermal growth factor receptor (EGFR) metabolic activity. Two hundred eighty-six unique locations nine samples freshly resected oral mucosa were imaged after topically applying EGF-Alexa 647 (to target EGFR) 2-(N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino)-2-deoxyglucose activity). Quantitative features calculated from resulting fluorescence images compared tissue histopathology maps. signal correlated well EGFR indicated by immunohistochemistry. A classification algorithm based on both resulted area under curve 0.83. Regions a posterior probability 0.80 1.00 contained more than 50% 99% (84/85) time. This study demonstrates how noninvasive used tool levels their across large characteristics correlate neoplasia. Applications include predicting regions highest likelihood disease, elucidating role biology, identifying patients who will respond
