摘要: In order to gain insight into the biological significance of a collagenase inhibitor secreted by human skin fibroblasts, we examined various connective tissues and body fluids for such protein. The inhibitors found in these were compared immunologically fibroblast Ouchterlony analysis development highly specific enzyme-linked immunosorbent assay (ELISA). Using this ELISA, cell cultures corneal gingival adult fetal lung fibroblasts similar amounts immunoreactive Each culture medium displayed reaction immunologic identity with when gel diffusion. testing functional inhibitory activity, same 1:1 stoichiometry inhibition was observed each that characterizes inhibitor. Other mesodermally derived types, including osteoblasts, uterine smooth muscle cells, fibrosarcoma explants tendon articular cartilage behaved manner as cultures. Because biochemical similarities have been described serum amniotic fluid, also sources proteins. data indicate both fluid contain which are functionally identical concentration serum, measured ELISA assay, is 1.03 +/- 0.27 micrograms/ml. results suggest equivalent synthesized many, if not all, mesodermal organism. apparently gains access certain fluid. This protein may, therefore, function regulation collagen degradation most tissues.
