摘要: Fourier transform infrared microscopy is a powerful tool for the characterization of mineral and protein in histologic sections bone. This study was concerned with determining whether techniques used to preserve these tissties prepare them sectioning had an effect on spectral properties. The υ1, υ3 phosphate bands 900–1200 cm-1 region were evaluate structure apatitic fresh-frozen, ethanol-fixed, formalin-fixed 35-day-old rat femurs; fresh-frozen 20-day-old fetal ground diaphyseal bone samples; formalin-fixed, methacrylate-embedded crystallinity (crystal size perfection) apatite assessed by curve-fitting analysis bands. Results indicate that ethanol or formalin fixation intact femur, embedding do not significantly alter apatite. However, did phase. In addition, evaluation secondary femur from Amide I II vibrations near 1650 1550 cm-1, respectively, revealed conformation altered fixation.
