Hypoglycemic effects of esculeoside A are mediated via activation of AMPK and upregulation of IRS-1

摘要: Tomato fruit (Lycopersicon esculentum Mill.) has been suggested to be useful for the prevention of diabetes. Esculeoside A is main saponin compounds in tomatoes. This study investigated hypoglycemic effects and underlying mechanism esculeoside C57BLKS/Leprdb (db/db) mice. Wild-type C57BLKS (db/dm) mice were used db/dm mouse group db/db randomly divided into 2 groups: untreated treated groups. (100 mg/kg) was administered by gavage 56 days group. Distilled water The blood liver biochemical parameters expression insulin signaling-related proteins examined. results showed that reduced fasting glucose (FBG) levels improved tolerance. Further investigation revealed hepatic protein expressions total AMP-activated kinase (T-AMPK), phosphorylated (p-AMPK), receptor substrate-1 (IRS-1), glucokinase (GCK) significantly upregulated after treatment. In contrast, phosphoenolpyruvate carboxykinase (PEPCK) downregulated These findings a potential alleviating metabolic abnormalities via regulation AMPK/IRS-1 pathway. Our supported possible application as functional supplement diabetes
