Effects of sulfonylureas on histochemical and ultracytochemical calcium distribution in B-cells of mice.

摘要: The study examines the effects of sulfonylurea compounds on histo- and ultracytochemical calcium distribution within B-cells mice using glyoxal-bis-(2-hydroxyanil) (GBHA) pyroantimonate method combined with X-ray microanalysis. Treatment tolbutamide (200 mg/kg), glibenclamide (2 mg/kg) glisoxepide causing moderate hypoglycemia B-cell degranulation, was associated an unchanged (30, 90 min) or slightly increased (180, 360 min, 4, 42 d) GBHA staining intensity islet cells compared controls. Ultra-cytochemically sulfonylureas provoked, controls, a redistribution calcium-rich, electron dense precipitates (EDPP). Precipitation predominantly occurred along inner surface plasma membranes granule halos. In contrast, cytoplasmic matrix, Golgi complexes rough endoplasmic reticulum contained only few fine precipitates. — investigated exerted identical in B-cells. results indicate that sulfonylurea-induced insulin secretion is accumulation This supports hypothesis altered handling by mainly accounts for insulinotropic effect sulfonylureas.
