摘要: Capillary electrophoresis-capacitively coupled contactless conductivity detection (CE-C4D), conducted using an in-house-developed polyvinyl alcohol (PVA)-coated capillary system, was applied for the simultaneous analysis of small anions and cations in saliva samples from wrestlers undergoing a weight training program. Use PVA CE provided good reproducible ion separation with minimization electroosmotic flow suppression protein adsorption onto wall. Four eight were separated 12min, background electrolyte 20mM MES/20mM histidine 18-crown-6 ether (pH 6) at 20kV. The relative standard deviations (n=5) migration times peak areas <1% <8%, respectively. limit signal-to-noise ratio 3 ranged 1.6 to 10μM. Using optimized CE-C4D we investigated correlations between concentrations salivary ions cortisol, which is commonly used as stress marker. Analysis ten wrestlers, who attempting rapid loss before competition, showed following trends: (1) all concentrations, except Ca(2+), Na(+), Cl(-), increased first last days loss; (2) Mg(2+) 166% (from 0.50mM 1.4mM) loss, being highest increase ions; (3) K(+), Mg(2+), NO3(-), SCN(-) levels strongly correlated (P<0.05) cortisol. rapidly produced useful data on contents, recovery determined by addition method (89-110%).
