摘要: This study reports the selection and characterization of osteogenic precursors from human bone marrow which were isolated by two “clonings” successive subculturing. These cell lines express alkaline phosphatase activity. Gel electrophoresis [3H]-proline labeled cultures showed that cloned cells produce only type I collagen. They synthetize osteocalcin osteonectin. respond to 1,25 dihydroxy vitamin D3 increasing synthesis secretion, parathyroid hormone cyclic AMP synthesis. After third subculture in absence β-glycerophosphate, these formed lots clusters exhibit high activity positive von Kossa staining. X-ray energy spectrum shows are surrounded “budding” structures containing calcium phosphorus with a ratio Ca:P identical those pure hydroxyapatite. process was associated with45Ca uptake into cells. All data support may be considerable clinical importance.
