摘要: Poly (2-hydroxyethylmethacrylate) (PolyHEMA) hydrogels, when combined with extracellular matrix molecules and infiltrated cultured Schwann cells, have the capability to induce CNS axonal regrowth after injury. We further investigated these PolyHEMA hydrogels their potential bridge injury sites. Collagen IV-impregnated containing cells were implanted into lesioned optic tract in 14 rats. On examination 2-4 months later, there was good adherence between implants tissue, large numbers of viable (S100+, GFAP+, Laminin+, LNGFR+) seen within hydrogel matrices. Immunohistochemical analysis showed that collagen preferentially supported transplanted not host glial such as astrocytes (GFAP+) or oligodendroglia (CAII+). Macrophages (ED1+) also sponge structure. Eighty-three percent contained RT97+ axons trabecular networks. Regrowing associated small number infiltrating astrocytes. traced up 510 microm from nearest neuropil. These sometimes myelinated by expressed peripheral myelin marker Po+. WGA/HRP-labeled retinal sponges, 40% cases growing for distances 350-450 polymer network. The data indicate impregnating sponges IV can modify reaction support survival cells. This study thus provides new information on how biomaterials could be used
