BRAFV600E mutation, but not RET/PTC rearrangements, is correlated with a lower expression of both thyroperoxidase and sodium iodide symporter genes in papillary thyroid cancer.

摘要: A low sodium iodide symporter (NIS) expression has been shown in papillary thyroid carcinomas (PTCs) harboring the BRAF V600E mutation. In present study, we analyzed mRNA of differentiation genes, glucose transporter (GLUT)-1 and GLUT-3, 78 PTCs according to presence or RET/PTC rearrangements. We found rearrangements 35.8 19.4% respectively. The NIS thyroperoxidase (TPO) genes were significantly lower (P<0.0001 P = 0.004 respectively) -positive PTC with respect non-mutated samples. support this result, immunohistochemistry showed that percentage NIS-positive cells was (P=0.005) -mutated (mean 53.5%) than negative cases 72.6%). contrast, no difference either any other genes' without When GLUT-1 GLUT-3 considered, correlation - nor RET/PTC-mutated cases. conclusion, study confirmed a genetic alteration and/or RET oncogenes 64% revealed significant mutation both TPO. This latterfinding could indicate an early dedifferentiation process is at molecular level PTC, thus suggesting previously demonstrated poor prognostic significance be related more advanced stage diagnosis.
