摘要: Monodisperse poly(glycidyl methacrylate–divinylbenzene) microspheres were functionalized with propyl sulfonic acid moieties to obtain beads negatively charged in a wide pH range. They packed into fused-silica capillary of 50 μm I.D. order separate proteins by electrochromatography (CEC). Baseline separation four basic as well three cytochrome c variants an average column efficiency 60 000 theoretical plates was obtained under isocratic elution conditions. The high is attributed the uniformity packing and hydrophilic surface coverage polymer derived from functionalization process. effect salt concentration on protein separations investigated results showed that CEC mechanism combination chromatographic retention electrophoretic migration. Moreover, strongly acidic also suitable for micro-HPLC gradient. comparison between two kinds modes shows described here exhibited higher peak than gradient micro-HPLC.
