摘要: Abstract A sensitive, specific and selective method has been developed for the simultaneous determination of bisoprolol hydrochlorothiazide in human plasma. The employed a state art LC–MS/MS operated positive negative ionization switching modes. simple sample preparation step involving protein precipitation with acetonitrile optimized; analytes internal standard moxifloxacin were separated on Purosphere ® STAR C 8 column (125 mm × 4 mm, 5 μm). mobile phase was an ammonium acetate solution (1 mM) formic acid (0.2%): methanol (65:17.5:17.5, v/v/v (%)), flow rate set at 0.65 mL/min. Bisoprolol ionized using ESI source prior to detection by Multiple Reaction Monitoring (MRM) mode while monitoring following transitions: m / z 326 → 116 bisoprolol, 296 → 269 296 → 205 hydrochlorothiazide. Linearity demonstrated over concentration range 0.10–30.0 (ng/mL) 1.00–80.00 ng/mL limits 0.100 1.00 validated successfully applied pharmacokinetic study 5 mg fumarate 12.5 mg tablet healthy volunteers.
