摘要: Fibronectin fragments (Fn-f), which are the breakdown products of fibronectin, accumulate in disc during degeneration and proved to induce intervertebral disc. The goal this investigation was determine functional role integrin α5β1, extracellular signal-regulated kinase (ERK), protein C (PKC) process Fn-f nucleus pulposus (NP) cells. We found that (100 nM, 30 kDa) exposure led NP cells, up-regulation α5β1 expression phosphorylation ERK1/2. After silenced ERK1/2 MMP9, MMP13, collagen II had no difference with control under treatment Fn-f. Finally, when inhibitor PKC were added into medium cells; we these two inhibitors could eliminate effect on It is concluded potential enhance cell a vicious circle. And subunit, ERK, all included loop. © 2010 Orthopaedic Research Society. Published by Wiley Periodicals, Inc. J Orthop Res 29:556–561, 2011
