摘要: Neuronal SNARE-proteins such as synaptobrevin, SNAP 25, and synaptotagmin are key players during neurosecretion. So far palmitoylation of SNAP-25 1 have been described in vivo. Here we analyzed the synaptobrevin 2 vitro using synaptosomal synaptic vesicle preparations from rat brain. Labeling vesicles prepared adult brain with [3H]palmitate revealed besides major palmitoylated proteins. [3H]Palmitoylation was resistant to chloroform/methanol extraction, but sensitive reducing agents indicating a covalent fatty acid bond cysteine residues. Palmitoylation also confirmed endogenous present PC-12 cells expressed vacciniavirus system Cos cells. In contrast situation seen membrane obtained brain, embryonic did not support [3H]palmitoylation synaptotagmin. These results suggest, that both were efficiently mature vesicles. However, at least one component machinery is developmentally upregulated.
