摘要: In the last few years, matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI) has been successfully used to study distribution of lipids within tissue sections. However, efforts have made acquire reliable quantitative data regarding localized concentrations these molecules. Here we propose an approach based on brain homogenates for quantification phosphatidylcholines (PCs) in section by MALDI MSI. Homogenates were spiked with a range PC(16:0 d31/18:1) concentrations. Sections from and intact simultaneously prepared before being analyzed MSI using Fourier transform ion cyclotron resonance (FT-ICR) analyzer. Standard curves generated signal intensity different ionic species ([M+H]+, [M+Na]+ [M+K]+) detected homogenate Localized finally extracted correlating standard intensities endogenous PC (especially PC(16:0/18:1)) areas section.
