摘要: Objective In equine recurrent uveitis (ERU), immune reactions are directed toward known antigens like S-antigen, interphotoreceptor retinoid-binding protein, and cellular retinalaldehyde-binding anti-retinal antibodies were detected in vitreous samples. The aim of this study was the investigation intraocular immunoglobulin M (IgM) reactivities to retinal proteome. Procedures Retina separated by one- two-dimensional gel electrophoresis blotted semidry on PVDF membranes. To identify IgM antibody responses tissue, blots incubated with samples ERU-diseased horses (n = 50) healthy controls (n = 30), followed an HRP-labeled secondary specific for IgM. Noticeable 2D western blot signals aligned a proteome, excised, subsequently identified tandem mass spectrometry. Results Interestingly, frequent very miscellaneous response patterns proteome 68% ERU detected. Binding localized at 17 different molecular weights. most frequently signal, 21 50 samples, located 49 kDa. Comparing interindividually between one up nine sample could be observed. All devoid antibodies. Analysis targeted spots spectrometry led clear identification 11 proteins (corresponding 16 spots). One candidate not discovered so far. Conclusion considerable demonstrates ongoing response, which might contribute remitting relapsing character ERU. Novel target point diverse pattern individual cases.
