摘要: Tissue plasminogen activator (tPA) activity in blood and plasma has been difficult to determine because it is unstable, especially subjects with high levels of inhibitor type 1 (PAI-1). We have attempted stabilize the tPA by collecting 9 volumes volume acidic citrate buffer which immediately lowers pH blood. At final 4.9 6.3, consistent were found. One composition, 0.5 mol/l 4.0, resulted 5.5, half-life 10 days an acceptably low degree haemolysis. This anticoagulant composition was selected for more extensive evaluation used collect samples from 29 volunteers morning after a 10-minute bed rest. Basal activity, mean SD, 0.47 +/- 0.21 IU/ml. After donating first sample, entered into 24-hour fast they donated second sample. During fast, basal increased 0.34 0.23 IU/ml (p = 0.05) PAI-1 decreased 11.4 11.6 6.3 8.9 U/ml 0.04).
