Cell Lines Derived from Human Parthenogenetic Embryos Can Display Aberrant Centriole Distribution and Altered Expression Levels of Mitotic Spindle Check-point Transcripts

摘要: Human parthenogenetic embryos have recently been proposed as an alternative, less controversial source of embryonic stem cell (ESC) lines; however many aspects related to the biology and derived lines still need be elucidated. We present here results on human (HP1 HP3) from blastocysts obtained by oocyte activation. Cell showed typical ESC morphology, expressed Oct-4, Nanog, Sox-2, Rex-1, alkaline phosphatase, SSEA-4, TRA 1-81 had high telomerase activity. Expression genes specific for different germ layers was detected HP cells differentiated upon embryoid body (EBs) formation. Furthermore, when cultured in appropriate conditions, were able differentiate into mature types neural hematopoietic lineages. However, injection undifferentiated immunodeficient mice resulted either poor differentiation or tumour formation with morphological characteristics myofibrosarcomas. Further analysis indicated aberrant levels molecules spindle well presence abnormal number centrioles autophagic Our confirm extend notion that can types, but also highlight possibility that, alteration proliferation mechanisms may occur these cells, suggesting great caution if a therapeutic use this kind is considered.
