α-spectrins are major ubiquitinated proteins in rat hippocampal neurons and components of ubiquitinated inclusions in neurodegenerative disorders

摘要: Abstract We have demonstrated that α-spectrins (αSpIΣ∗ and αSpIIΣ1) are major ubiquitinated proteins in terminally differentiated hippocampal neurons culture. Western blotting experiments, using αSpIΣ1, αSpIIΣ1, ubiquitin antibodies lysates of 11-day-old cultured rat neurons, a single band comigrating with αSpIΣ∗ αSpIIΣ1 5% polyacrylamide sodium dodecyl sulfate gel is recognized by when 125 I-protein A used for detection. Immunofluorescence staining the 7- 12 -day-old neuron cultures ubiquitin, all these label but not astrocytes cultures. Immunoprecipitation spectrin subunits 12-day-old under stringent conditions (9.5 M urea) αSpIΣ1 followed blot experiments immunoprecipitated confirmed both neurons.
