摘要: Abstract Aims Matrine (MAT), a quinolizidine alkaloid derived from the herb Radix Sophorae flavescens , has been recently found to be beneficial in experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis, mainly through its anti-inflammatory effect. In present study, we tested effect MAT on ongoing EAE and defined possible mechanisms underlying effects myelination oligodendrocytes. Main methods was induced C57BL/6 mice treatment started at disease onset. Clinical scores were monitored daily; spinal cords corpus callosum brain region harvested day 23 p.i. for inflammatory infiltration demyelination central nervous system. Myelin content development oligodendrocytes their precursors determined by immunostaining, expression p-Akt, p-mTOR, p-PI3K, p-P70S6 Western blot. Key findings effectively suppressed severity increased proteolipid protein, myelin protein that is marker CNS myelin. largely number mature oligodendrocytes, significantly activated PI3K/Akt/mTOR signaling pathway, which required oligodendrocyte survival axon myelination. Significance These demonstrate differentiation during EAE, most likely activating pathway.
