Activation of phospholipase D in CHO cells transfected with the human epidermal growth factor (EGF) receptor: Differential effects of protein kinase C activation and EGF

摘要: Multiple intracellular signal transduction pathways, including phospholipases A2 and D, can be activated by epidermal growth factor (EGF) in both a protein kinase C (PKC)-dependent -independent manner. We investigated the activation of phospholipase D (PLD) PKC activator, phorbol myristate acetate (PMA) EGF CHO cells transfected with full-length receptor. In labelled arachidonic acid or linoleic acid, PMA PLD, determined formation transphosphatidylation product phosphatidylethanol presence ethanol. A basal PLD activity was seen acid-labelled but not acid. This augmented phosphotyrosine phosphatase inhibitor vanadate reduced tyrosine inhibition contributed to PKC, as could elicited following prolonged exposure ester, known down-regulate some isoforms. By contrast, failed stimulate either fatty species. It is proposed that condition PKC-dependent phosphorylation are linked (possibly (PLC)-mediated diacylglycerol); which (PLA2) elicit PLC these without further effect on PLD.
