Strategies for analysis of electrophoretically separated proteins and peptides

摘要: Abstract Electrophoresis methodology is unsurpassed in separating individual proteins and peptides from a mixture regarded by the majority of protein biochemists as choice. Proteins can be eluted into solution or electroblotted onto membrane subjected to direct sequencing, amino acid composition post-translational modification analyses. Applications highly reproducible 2D electrophoresis, “in-gel” “on-blot” fragmentation procedures, high sensitivity N-terminal sequencing variety mass spectrometric methods (post-source decay MALDI–MS, LC–ESI MS/MS CE–MS) provide invaluable tools for characterisation recombinant identification unknown proteins. In this article we overview strategies employed recent years analysis separated various electrophoretic techniques. Methodological approach used newly emerged virus presented an example effective strategy.
