Loss of TIP1;1 aquaporin in Arabidopsis leads to cell and plant death

摘要: Arabidopsis TIP1;1 (gammaTIP) is a member of the tonoplast family aquaporins (AQP). Using RNA interference (RNAi) we reduced to different extent in various lines. When most severely affected, miniature plants died, phenotype partially complemented by homolog McMIP-F. Less affected lines produced small plants, early senescence, and showed lesion formation. The relative water content RNAi was not significantly affected. Global expression profiling suggested disturbance carbon metabolism with upregulated transcripts for functions acquisition respiration, vesicle transport, signaling transcription, radical oxygen stress. Metabolite profiles low glucose, fructose, inositol, threonic, succinic, fumaric, malic acids, but sucrose levels were similar WT. Increased amounts found raffinose several unknown compounds. also contained high starch apoplastic carbohydrate increased. A GFP-TIP1;1 fusion protein indicated location spongy mesophyll cells, signal intensity palisade associated vesicles near plastids. Signals vascular tissues strongest only vesicle-like structures outlined large vacuoles. Compromised routing lack provision cell-autonomous seems characterize this phenotype. We suggest function vesicle-based metabolite through or between pre-vacuolar compartments central vacuole. Phenotype characteristics support view functioning as marker that are targeted
