Topotecan Lactone Selectively Binds to Double- and Single-Stranded DNA in the Absence of Topoisomerase I

摘要: We report the first experimental observation that a clinically important camptothecin [CPT; topotecan (TPT), water-soluble CPT] binds directly and noncovalently to double-stranded DNA single-stranded structures in absence of topoisomerase I, but only lactone form. observed clear sequence specificity TPT binding duplex DNA, which was comprised alternating purine-pyrimidine sequences contained dT. These structural studies direct lactone-DNA support several considerations involving possible mechanism(s) anticancer activity CPT-type drugs containing 20( S ) moiety.
