Hepatotoxin-induced hypertaurinuria: a proton NMR study

摘要: The urinary excretion of taurine by rats after dosing with various hepatotoxins has been investigated by1H NMR spectroscopy. After single hepatotoxic doses hydrazine, carbon tetrachloride, 1-naphthylisothiocyanate, or thioacetamide there was biochemical and histopathological evidence hepatic damage. Proton spectroscopy the urine collected for 24 h from these animals revealed a marked elevation in (control 11.9 μmole/h/kg) (42.2 μmole/h/kg), tetrachloride (52.5 gmmole/ h/kg), 1-naphthyl-isothiocyanate (80.4 hydrazine (52.9 μmmole/h/kg). allyl alcohol administration no increase (7.5 μmol/h/kg). dose related. High resolution proton spectroscopic analysis also resonances several metabolites an N-acetylcysteine conjugate alcohol, acetamide as metabolite respective compounds. Changes endogenous substances that may be related to pathological events were detected, such decrease 2-oxoglutarate citrate both administration. results confirm is powerful analytical tool evaluation study toxic substances. Furthermore, measurement provide non-invasive indicator acute damage certain classes hepatotoxins.
