摘要: Requirements for the antimicrobial activity of an antibiotic are: (1) binding drug to a specific site in bacteria; (2) occupation critical number sites; and (3) persistence at these sites sufficient time. With concentration-dependent antibiotics ratio peak serum concentration MIC pathogen is primary determinant bacterial killing; with concentration-independent it length time remains above MIC, rather than level. The pharmacokinetics new macrolides azithromycin clarithromycin differ notably from those conventional more rapid extensive distribution body tissues. Because unique tissue pharmacokinetics, pharmacodynamic models that apply other classes may not explain clinical efficacy clarithromycin.
