摘要: Abstract Histamine produces a rapid and massive increase of the c-GMP level guinea-pig lung tissue. The ec 50 value for this in vitro response is found to be 27 μM maximally 9-fold elevated by 100 histamine. stereoselectively inhibited enantiomers chlorpheniramine, indicating H1-receptor involvement. Preincubation tissue with 200 NCDC, phospholipase C inhibitor, reduces histamine (100 μM) responses 16 ± 3% (N = 6) control production. Inhibition protein kinase H-7 does not significantly attenuate response, whereas omittance extracellular Ca2+ results almost complete inhibition histamine-induced hemoglobin, methylene blue antioxidants butylated hydroxytoluene nordihydroguaretic acid, involvement nitric oxide-dependent activation soluble guanylate cyclase. This suggestion supported concentration-dependent production NG-monomethyl- l -arginine (NMA). At concentration 20 NMA 34 8% response. reversed 127 20% exogenous addition 1 mM -arginine. These findings show that after an initial H1-receptor-mediated, C-dependent, Ca2+-mobilization enzymatic conversion oxide stimulated. finally responsible cyclase, leading c-GMP.
