摘要: OBJECTIVES To assess the presence of a local angiotensin-generating systems (LAGS) and its participation in tumor growth human pancreatic cancer derived cell line Capan-1. METHODS Capan-1 cells were cultured Dulbecco modified Eagle medium, angiotensin I was assayed by radioimmunoassay II vascular endothelial factor enzyme-linked immunosorbent assay supernatant. Immunohistochemistry reverse transcription-polymerase chain reaction performed for expression AT1 AT2 receptors. Angiotensin binding assays blockade studied. RESULTS High levels both angiotensins found cells, although neither nor detected culture Reverse immunocytochemistry revealed that do not express receptors; however, specific to membrane identified II. Neither exogenous Dup753 (specific receptor blocker) affected cells' proliferation or secretion. CONCLUSIONS Detection along with provides evidence existence LAGS operates an intracrine manner. Intracellular may play role aggressiveness is possible target therapeutic agents.
