Quantitative Analysis of Bacitracin in Porcine Edible Tissues by High-Performance Liquid Chromatography–Electrospray Ionization Tandem Mass Spectrometry and Its Application to Residue Depletion Study

摘要: The residue kinetics of bacitracin in animals has rarely been quantitatively described due to inadequate sensitivity analytical methods. A sensitive and selective quantitative liquid chromatography coupled with electrospray ionization tandem mass spectrometry (LC-MS/MS) method was developed determine the depletion zinc pigs. tissues samples were extracted acidified methanol/water solution (v/v, 1:1, 1 % formic acid) followed by liquid-liquid extraction saturated 0.1 mmol dithizone chloroform. extracts subjected Phenomenex Strata-X cartridges for further cleanup 0.1 % acid methanol. Analysis performed on a C18 column positive-ion multiple-reaction monitoring mode. product ions at m/z 199 227 selected confirmation quantification. linear concentration ranges 150–4000 μg/L limit detection (LOD) quantification (LOQ) 25 50 μg/kg kidney 20 skin fat, liver, muscle. recoveries from different higher than 82.1 % three spiking levels relative standard deviations (RSDs) less 8.8 %. study showed that longer time detected other tissues. elimination half-life 0.45 days, recommended withdrawal period zero. These results suggest established satisfactorily enables closer swine.
