Optimization of high-speed counter-current chromatography for separation of polyphenols from the extract of hawthorn (Crataegus laevigata) with response surface methodology

摘要: Abstract Separation and purification of polyphenols from the extract hawthorn ( Crataegus laevigata ) was investigated with high-speed counter-current chromatography (HSCCC). Solvent systems were examined according to partition coefficients target components n-butanol–ethyl acetate–water (1:1:2, v/v/v) system chosen. The critical experimental parameters HSCCC optimized determined response surface methodology (RSM) as following: Revolution speed 850 rpm, flow-rate 1.5 mL/min temperature 25 °C. Under these conditions, 500 mg crude introduced into a product 9.7 mg purity over 94.8% obtained in 260 min, which identified chlorogenic acid UV, LC–MS 1 H NMR. result proved that RSM is an efficient method optimize could be very effective approach separate purify natural source.
