摘要: Advanced platelet-rich fibrin (A-PRF) is an autogenous biological material obtained from peripheral blood. A-PRF extract (A-PRFe) contains a high concentration of various cytokines that are increasingly appreciated for their roles in improving stem cell repairing function during tissue regeneration. However, the optimal A-PRFe to stimulate cells unknown. This study aimed identify concentrations promote adipogenic and osteogenic differentiation human adipose-derived (ASCs). We produced clots by centrifuging fresh blood samples isolated identified ASCs using surface CD markers multilineage potential. Enzyme-linked immunosorbent assay (ELISA) showed several cytokines, including b-FGF, PDGF-BB, others, increased gradually, peaked on day 7 then decreased. Cell proliferation assays significantly stimulated ASC proliferation, at higher doses. The degree culture medium, as determined oil red O alizarin staining. Reverse transcription polymerase chain reaction (RT-PCR) expression levels genes related adipogenic/osteogenic (PPARγ2, C/EBPα, FABP4, Adiponectin, ALP, OPN, OCN, RUNX2), paracrine (HIF-1α, VEGF, IGF-2) immunoregulation (HSP70, IL-8) were groups with than lower groups. demonstrates ideal use applications regenerative medicine because it improves functions, differentiation, dose-dependent manner.
