MiR-21 Suppresses Anoikis through Targeting PDCD4 and PTEN in Human Esophageal Adenocarcinoma

摘要: Anoikis is a form of apoptosis induced upon cell detachment from extracellular matrix. It has been determined that acquisition resistance to anoikis critical step for tumor metastasis. MiR-21, the most prominent oncomiR, plays an important role in progression. In this study, we revealed up-regulation miR-21 human esophageal adenocarcinoma (EA) associated with lymph node metastasis and poor survival rate. Because established anti-apoptosis effect miR-21, it tempting speculate might contribute by regulating anoikis. qRT-PCR analysis demonstrated expression OE33/AR cells (subpopulation EA OE33 acquired anoikis) was significantly increased. Also, transfection mimics provided resisting By luciferase assays, verified PDCD4 PTEN were functional targets miR-21. mouse model, via tail vein injection experiment, showed formation vivo could be mediated changing pattern.
