Mutually exclusive sense–antisense transcription at FLC facilitates environmentally induced gene repression

摘要: Antisense transcription through genic regions is pervasive in most genomes; however, its functional significance still unclear. We are studying the role of antisense transcripts (COOLAIR) cold-induced, epigenetic silencing Arabidopsis FLOWERING LOCUS C (FLC), a regulator transition to reproduction. Here we use single-molecule RNA FISH address mechanistic relationship FLC and COOLAIR at cellular level. demonstrate that while sense can co-occur same cell they mutually exclusive individual loci. Cold strongly upregulates an increased number cells facilitates shutdown cis. form dense clouds each locus, acting influence changed H3K36me3 dynamics. These results may have general implications for other loci showing both transcription.
